Influenza A virus of subtypes H5 and H7 is detected using RT PCR, whereby cDNA is synthesized from RNA and its specific fragment is amplified millions of times, which allows visualization of the results using agarose gel electrophoresis.
The test system is designed so that only the subtypes H5 and H7 are detected in the course of RT PCR. The differentiation between the two subtypes is accomplished in the course of reamplification, when two specific pairs of primers are used for each subtype. The resulting PCR fragments differ in length, which further facilitates the differentiation.
